In 2023, prospection of a dried-out lake near Papowo Biskupie in north-central Poland identified substantial deposits of bronze artefacts. Excavation revealed further deposits and dozens of human skeletons that date from 1000–400 BC, suggesting that the site held particular significance as a place for sacrificial offerings in the Lusatian culture.

Introduction
Hereditary Transthyretin amyloidosis (hATTR) is a rare autosomal dominant, multisystemic, progressive, and potentially fatal genetic disease. Currently, the only drug made available in the Brazilian National Health System to treat hATTR is tafamidis meglumine, indicated for symptomatic adult patients in early stage (stage 1) and not undergoing liver transplantation for amyloidosis associated with hATTR.
MethodsA systematic review was conducted in the databases MEDLINE via Pubmed, Embase, The Cochrane Library, and LILACS addressing the question “Is patisiran treatment effective and safe for patients diagnosed with ATTRh amyloidosis with stage 2 polyneuropathy or who have an inadequate response to tafamidis?”
ResultsThe 13 studies included in the review demonstrate the efficacy of patisiran in reducing the neuropathic progression of the disease, as evidenced by decreased mNIS+7 scale scores following 18-month use of the drug. Improvements in the quality of life of patients taking patisiran have been reported, as measured by reduced scores on the Norfolk-QoL-DN scale. Patisiran has also been shown to be effective in reducing NT-proBNP, a marker related to cardiac stress. Improvements in the nutritional status of patients taking patisiran were demonstrated by increasing modified body mass index (BMI). Good tolerability of patisiran was observed by patients using it. Most adverse events were classified as mild or moderate. The studies indicated that the occurrence of deaths is similar between the patisiran and placebo groups. Most deaths were related to cardiac events and were not associated with the use of patisiran.
ConclusionsThe use of patisiran in patients with hATTR demonstrated efficacy in reducing the neuropathic progression of the disease, evidenced by decreased mNIS+7 scale scores, improvements in quality of life as measured by reduced Norfolk-QoL-D scale scores, and reduced NT-proBNP. The drug patisiran was well tolerated, with most adverse events rated as mild and moderate.

Mathematical models of polyelectrolyte gels are often simplified by assuming the gel is electrically neutral. The rationale behind this assumption is that the thickness of the electric double layer (EDL) at the free surface of the gel is small compared to the size of the gel. Hence, the thin-EDL limit is taken, in which the thickness of the EDL is set to zero. Despite the widespread use of the thin-EDL limit, the solutions in the EDL are rarely computed and shown to match to the solutions for the electrically neutral bulk. The aims of this paper are to study the structure of the EDL and establish the validity of the thin-EDL limit. The model for the gel accounts for phase separation, which gives rise to diffuse interfaces with a thickness described by the Kuhn length. We show that the solutions in the EDL can only be asymptotically matched to the solutions for an electrically neutral bulk, in general, when the Debye length is much smaller than the Kuhn length. If the Debye length is similar to or larger than the Kuhn length, then phase separation can be initiated in the EDL. This phase separation spreads into the bulk of the gel and gives rise to electrically charged layers with different degrees of swelling. Thus, the thin-EDL limit and the assumption of electroneutrality only generally apply when the Debye length is much smaller than the Kuhn length.
The effect of a prospective intervention program with automated monitoring of hand hygiene performance in long-term and acute-care units at a Veterans Affairs medical center

Objective:
To measure the impact of an automated hand hygiene monitoring system (AHHMS) and an intervention program of complementary strategies on hand hygiene (HH) performance in both acute-care and long-term care (LTC) units.
Design:Prospective, nonrandomized, before-and-after intervention study.
Setting:Single Veterans Affairs Medical Center (VAMC), with 2 acute-care units and 6 LTC units.
Methods:An AHHMS that provides group HH performance rates was implemented on 8 units at a VAMC from March 2021 through April 2022. After a 4-week baseline period and 2.5-week washout period, the 52-week intervention period included multiple evidence-based components designed to improve HH compliance. Unit HH performance rates were expressed as the number of dispenses (events) divided by the number of patient room entries and exits (opportunities) × 100. Statistical analysis was performed with a Poisson general additive mixed model.
Results:During the 4-week baseline period, the median HH performance rate was 18.6 (95% CI, 16.5–21.0) for all 8 units. During the intervention period, the median HH rate increased to 21.6 (95% CI, 19.1–24.4; P < .0001), and during the last 4 weeks of the intervention period (exactly 1 year after baseline), the 8 units exhibited a median HH rate of 25.1 (95% CI, 22.2–28.4; P < .0001). The median HH rate increased from 17.5 to 20.0 (P < .0001) in LTC units and from 22.9 to 27.2 (P < .0001) in acute-care units.
Conclusions:The intervention was associated with increased HH performance rates for all units. The performance of acute-care units was consistently higher than LTC units, which have more visitors and more mobile veterans.

A new two-phase model for concentrated suspensions is derived that incorporates a constitutive law combining the rheology for non-Brownian suspension and granular flow. The resulting model exhibits a yield-stress behaviour for the solid phase depending on the collision pressure. This property is investigated for the simple geometry of plane Poiseuille flow, where an unyielded or jammed zone of finite width arises in the centre of the channel. For the steady states of this problem, the governing equations are reduced to a boundary value problem for a system of ordinary differential equations and the conditions for existence of solutions with jammed regions are investigated using phase-space methods. For the general time-dependent case a new drift-flux model is derived using matched asymptotic expansions that takes into account the boundary layers at the walls and the interface between the yielded and unyielded region. The drift-flux model is used to numerically study the dynamic behaviour of the suspension flow, including the appearance and evolution of an unyielded or jammed regions.

Ca plays an essential role in bone development; however, little is known about its effect on intestinal gene expression in juvenile animals. In the present study, thirty-two weaned pigs (9·5 (sem 0·11) kg) were assigned to four diets that differed in Ca concentration (adequate v. high) and cereal composition (wheat–barley v. maize) to assess the jejunal and colonic gene expression of nutrient transporters, tight junction proteins, cytokines and pathogen-associated molecular patterns, nutrient digestibility, Ca balance and serum acute-phase response. To estimate the impact of mucosal bacteria on colonic gene expression, Spearman's correlations between colonic gene expression and bacterial abundance were computed. Faecal Ca excretion indicated that more Ca was available along the intestinal tract of the pigs fed high Ca diets as compared to the pigs fed adequate Ca diets (P< 0·05). High Ca diets decreased jejunal zonula occludens 1 (ZO1) and occludin (OCLN) expression, up-regulated jejunal expression of toll-like receptor 2 (TLR2) and down-regulated colonic GLUT2 expression as compared to the adequate Ca diets (P< 0·05). Dietary cereal composition up-regulated jejunal TLR2 expression and interacted (P= 0·021) with dietary Ca on colonic IL1B expression; high Ca concentration up-regulated IL1B expression with wheat–barley diets and down-regulated it with maize diets. Spearman's correlations (r>0·35; P< 0·05) indicated an association between operational taxonomic units assigned to the phyla Bacteroidetes, Firmicutes and Proteobacteria and bacterial metabolites and mucosal gene expression in the colon. The present results indicate that high Ca diets have the potential to modify the jejunal and colonic mucosal gene expression response which, in turn, interacts with the composition of the basal diet and mucosa-associated bacteria in weaned pigs.
Mesophotic surveys of the flora and fauna at Johnston Atoll, Central Pacific Ocean

Despite its extreme geographical isolation, numerous expeditions have surveyed the marine flora and fauna of Johnston Atoll. However, historical information about the marine biodiversity of Johnston is mostly limited to SCUBA surveys in shallow-waters (<30 m), and submersible observations in deeper waters (100–500 m). Extensive coral reefs, known as mesophotic coral ecosystems, exist between these two depth ranges at Johnston, but have remained largely unexplored. We used closed-circuit rebreathers to survey eleven sites at mesophotic depths (32–78 m) surrounding Johnston Atoll. A total of 130 species were recorded, including 99 species of fish, 15 species of corals, nine species of macroalgae, three species of echinoderms, three species of sponges and one species of squat lobster. Most species recorded during our mesophotic surveys have previously been recorded on shallow-water (<30 m) reefs at Johnston, with the exception of one black coral, one zoanthid, one squat lobster, two macroalgae, three sponges, and 22 fish, which represent new records for the atoll. As noted in previous studies, our surveys found a near absence of endemism, and recorded high proportions of species that are also known from the Hawaiian Archipelago. The similarity between the mesophotic biodiversity of Johnston Atoll and Hawaiʻi provides further support for the strong connectivity between these two locations highlighted in previous studies.
Developing risk hypotheses and selecting species for assessing non-target impacts of GM trees with novel traits: The case of altered-lignin pine trees

A procedure is presented for developing environmental risk hypotheses associated with the deployment of forest trees genetically modified to have altered wood properties and for selecting non-target species to test these hypotheses. Altered-lignin Pinus radiata trees intended for use in New Zealand are used as a hypothetical case study to illustrate our approach. Firstly, environmental management goals (such as wood production, flood control or preservation of biodiversity) were identified and linked to the forest attributes they require. Necessary conditions for each attribute were listed and appropriate assessment endpoints for them developed. For example, biological control of pests may be one condition necessary for a forest to have healthy trees, and the diversity and abundance of natural enemy species in the forest could be an appropriate assessment endpoint for measuring this condition. A conceptual model describing the relationships between an altered-lignin GM pine tree and potentially affected invertebrates and micro-organisms in a plantation forest was used to develop a set of risk hypotheses describing how the GM trees might affect each assessment endpoint. Because purified lignin does not represent the properties it imparts to wood, maximum hazard dose tests with non-target organisms, as are used to inform toxin risk assessment, cannot be conducted. Alternative experiments, based on current knowledge of the responses of organisms to lignin, must be designed. A screening method was adapted and applied to a database of invertebrate species known to inhabit New Zealand pine forests to identify and prioritize non-target invertebrate species that could be used as experimental subjects for examining these hypotheses. The screening model and its application are presented, along with a set of recommendations for pre-release tests with GM pines and potentially affected invertebrates and micro-organisms.

Antioxidant requirements have neither been defined for endurance nor been defined for ultra-endurance athletes. To verify whether an acute bout of ultra-endurance exercise modifies the need for nutritive antioxidants, we aimed (1) to investigate the changes of endogenous and exogenous antioxidants in response to an Ironman triathlon; (2) to particularise the relevance of antioxidant responses to the indices of oxidatively damaged blood lipids, blood cell compounds and lymphocyte DNA and (3) to examine whether potential time-points of increased susceptibility to oxidative damage are associated with alterations in the antioxidant status. Blood that was collected from forty-two well-trained male athletes 2 d pre-race, immediately post-race, and 1, 5 and 19 d later was sampled. The key findings of the present study are as follows: (1) Immediately post-race, vitamin C, α-tocopherol, and levels of the Trolox equivalent antioxidant capacity, the ferric reducing ability of plasma and the oxygen radical absorbance capacity (ORAC) assays increased significantly. Exercise-induced changes in the plasma antioxidant capacity were associated with changes in uric acid, bilirubin and vitamin C. (2) Significant inverse correlations between ORAC levels and indices of oxidatively damaged DNA immediately and 1 d post-race suggest a protective role of the acute antioxidant responses in DNA stability. (3) Significant decreases in carotenoids and γ-tocopherol 1 d post-race indicate that the antioxidant intake during the first 24 h of recovery following an acute ultra-endurance exercise requires specific attention. Furthermore, the present study illustrates the importance of a diversified and well-balanced diet to maintain a physiological antioxidant status in ultra-endurance athletes in reference to recommendations.

We derive the asymptotic form of the self-similar solutions of the second kind of the Cauchy problem for Barenblatt's nonlinear filtration equation by perturbing the Lie group of the underlying linear problem. We also show that the decay rate, appearing in the similarity solutions, can be found by a simple inspection of the corresponding energy dissipation law.

Summary
INTRODUCTION
Pseudomonas aeruginosa is a ubiquitous Gram-negative microorganism that thrives in many environments, from soil and water to animals and people. It is an opportunistic pathogen that can cause respiratory infections, urinary tract infections, gastrointestinal infections, keratitis, otitis media, and bacteremia. P. aeruginosa is the fourth most common nosocomial pathogen, accounting for approximately 10% of hospital-acquired infections (www.cdc.gov). Immunocompromised patients, such as those undergoing cancer treatment or those infected with AIDS, burn patients, or cystic fibrosis (CF) patients, are susceptible to P. aeruginosa infections. These infections are difficult to treat by using conventional antibiotic therapies, and hence result in significant morbidity and mortality in such patients. The recalcitrant nature of P. aeruginosa infections is thought to be due to the organism's intrinsic antibiotic resistance mechanisms and its ability to form communities of bacteria encased in an exopolysaccharide matrix; such communities are known as biofilms.
P. aeruginosa possesses an impressive arsenal of virulence factors to initiate infection and persist in the host. These include secreted factors, such as elastase, proteases, phospholipase C, hydrogen cyanide, exotoxin A, and exoenzyme S, as well as cell-associated factors, such as lipopolysaccharide (LPS), flagella, and pili. The expression of these factors is tightly regulated. Many factors are expressed in a cell-density-dependent manner known as quorum sensing (QS). Quorum sensing, or cell-to-cell communication, is a means by which bacteria can monitor cell density and coordinate population behavior. The behavior was first identified in Vibrio fischeri as a mechanism to induce bioluminescence (20).
Involuntary out-patient commitment and reduction of violent behaviour in persons with severe mental illness

Background
Violent behaviour among persons with severe mental illness (SMI) causes public concern and is associated with illness relapse, hospital recidivism and poor outcomes in community-based treatment.
AimsTo test whether involuntary out-patient commitment (OPC) may help to reduce the incidence of violence among persons with SMI.
MethodOne-year randomised trial of the effectiveness of OPC in 262 subjects with psychotic or major mood disorders and a history of hospital recidivism. Involuntarily hospitalised subjects awaiting OPC were randomly assigned to release or court-ordered treatment after discharge. Those with a recent history of serious assault remained under OPC until expiry of the court order (up to 90 days); then OPC orders were renewed at clinical/court discretion. Control subjects had no OPC. Four-monthly follow-up interviews with subject, case manager and collateral informant took place and service records were collected.
ResultsA significantly lower incidence of violent behaviour occurred in subjects with ⩾6 months' OPC. Lowest risk of violence was associated with extended OPC combined with regular out-patient services, adherence to prescribed medications and no substance misuse.
ConclusionsOPC may significantly reduce risk of violent behaviour in persons with SMI, in part by improving adherence to medications while diminishing substance misuse.
